No idea about the numbers, but yes, you can use a big population to keep a good amount of diversity. Usually you do selection on a landrace, trying to get a population fit for your conditions, and that also meat your goals. In other words, you cull most of what doesn't grow well for you, and what doesn't fit your selection criteria.
You could select with diversity in mind, taking seed from as many plants as possible, and making sure of taking seed of those that look different. You'll loose some genes, dilute others, but most will stay there somewhere.
